An organic-inorganic composite scaffold was designed for peripheral nerve regeneration. RGD Arg-Gly-Asp grafted poly{lactic acid–co- glycolic acid-alt-L-lysine} PRGD was synthesize to promote nerve regeneration for its inducing effect on nerve cells adhesion and guidance on directional growth of axons. Inorganic nano hydroxyapatite n-HAP was induced into polymer matrix to neutralize the acidity produced by the degradation products and supply Ca2+ and PO4 3- ions for axon regeneration and protein tyrosine phosphorylation. The porosity of the obtained PRGD-PDLLA-n-HAP scaffold was about 98%. Its hydrophilicity and cell affinity were improved by n-HAP. The added HAP nanoparticles can promote the RSC96 Schwann cells adhesion, proliferation and growth both on the surface of materials and inside the scaffold.
